what do I do with C,D, and E drives

For starters, you can move all of your data files from C:\ (which is probably getting pretty full) to the unused partition. Additionally, when you install new programs, make a Program Files folder on D:\ and install your new programs onto it. If you need room on C:\ you can also uninstall programs currently in C:\Program Files and reinstall them in D:\Program Files.

Another thing I do with my other partitions is save all of my downloads in a "Downloaded .exe" folder. In case I should have to format and reinstall my operating system in C:\ I can simply install from the downloaded files on E:\ (in my case) without having to download them again. Since I am restricted to a dial-up connection, this represents a tremendous time saver.
